Klaassen received her training with the artist Gijs Jacobs van den Hof, who in 1928 recommended that she continue her studies with Jan Bronner at the Rijksakademie van beeldende kunsten in Amsterdam. She completed her training in 1932, in which year she won the prestigious Prix de Rome for monumental and decorative sculpture. She was a member of the artist association St. Luke's and the Dutch Circle of Sculptors.
In 1948 she produced the Inauguration Medal to mark the inauguration on Sept. 6, 1948, of Queen Juliana. She and several other artists were involved in the interior decorations of the passenger ships SS Nieuw Amsterdam (II) (parquet intarsias smoking room in 1937) and Rotterdam (V) (1959) for the Holland-America Line, as well as Willem Ruys (parquet intarsias for public rooms in 1938) for the Royal Rotterdam Lloyd.
Nel Klaassen worked as a sculptor on the construction of the Rotterdam Schouwburg (1945 - 1947), which has since been demolished, the bank buildings of the Nederlandsche Handel-Maatschappij in Amsterdam and Rotterdam (1950), the Amsterdamsche Bank in Rotterdam (1950) and the department store Magazijn de Bijenkorf in The Hague.
